In the present paper we have compared the time course of inactivation of the Rnase H function of E. coli DNA polymerase III holoenzyme by histidine 2 as a substrate and by NH2OH. The rate of inactivation was similar, indicating that histidine 2 is not the catalytic base for the RNA hydrolysis reaction. We have also studied the effect of alkylation of lysine 32, which shows that this residue is an essential component of the catalytic mechanism of the enzyme. In the presence of saturating concentrations of dATP, dTTP and dGTP, lysine 32 alkylation results in a marked decrease in the amount of the active holoenzyme with reduced RNA polymerase activity.
